Immunomodulation of Potent Antioxidant Agents: Preclinical Study to Clinical Application in Multiple Sclerosis
نویسندگان
چکیده
Multiple sclerosis (MS) is a chronic disease of the central nerve system (CNS) primarily affecting youngsters. The CNS has a potent antioxidant defense mechanism to scavenge reactive oxygen species (ROS). MS can be effectively studied in animal models with experimental autoimmune encephalomyelitis (EAE). The immunopathogenic mechanisms involved in EAE are attributed to T-cell-mediated inflammatory disease of the CNS and activation of T cells, which recruit invading inflammatory cells, such as macrophages. The activated T cells also induce astrocytes and microglia in situ to secrete cytokines, chemokines, and toxic molecules, namely, glutamate, nitric oxide (NO), and/or ROS, which in turn contribute to axonal damage. These processes are followed by complement activation and antibody-mediated phagocytosis of axons, which eventually lead to demyelination and axonal injury.

O 9: Immunomodulatory Effects of Neural Stem Cell on Multiple Sclerosis: A Systematic Review
Multiple sclerosis (MS) and its animal model, experimental autoimmune encephalomyelitis (EAE), are chronic inflammatory demyelinating disorders of central nervous system (CNS). While the cause is unclear, the fundamental mechanism is thought to be destruction of myelin sheaths of neurons through immune system. One of the approaches being proposed in EAE therapy is neural stem cells (NSCs) trans...
متن کاملThe Role of Stem Cell Therapy in Multiple Sclerosis: an Overview of the Current Status of the Clinical Studies
The complexity of multiple sclerosis (MS) and the incompetence of a large number of promise treatments in MS urge us to plan new and more effective therapeutic approaches that aim to suppress ongoing autoimmune responses and induction of local endogenous regeneration. Emerging data propose that hematopoietic, mesenchymal and neural stem cells have the potential to restore self-tolerance, to pro...
متن کاملInterferon β-1a and Atorvastatin in the Treatment of Multiple Sclerosis
Background: Statins, widely used cholesterol-lowering agents, have also been demonstrated to have anti-inflammatory and immunomdulatory effects. Objective: To evaluate the effects of atorvastatin in combination with Interferon-β in the treatment of multiple sclerosis (MS) in a randomized controlled clinical trial. Methods: Multiple sclerosis patients were randomized independently, in a double b...
متن کاملPathophysiological Concepts in Multiple Sclerosis and the Therapeutic Effects of Hydrogen Sulfide
Introduction: Multiple sclerosis (MS) is generally known as a manageable but not yet curable autoimmune disease affecting central nervous system. A potential therapeutic approach should possess several properties: Prevent immune system from damaging the brain and spinal cord, promote differentiation of oligodendrocyte progenitor cells (OPCs) into mature oligodendrocytes to p...
متن کاملSystematic review: Superparamagnetic Iron Oxide nanoparticles as contrast agents in diagnosis of multiple sclerosis
Several MRI contrast agents (CAs) are used in medical diagnosis that gadolinium (Gd3+) is the most widely used as contrast agents. Unfortunately, its toxicity is due to its inefficiency. In this review, we discuss about the ability of SPIONs in MRI and application in Multiple Sclerosis diagnosis. Superparamagnetic iron oxide nanoparticles (SPIONs) such as magnetite nanoparticles are used as goo...
